چکیده

Background Host immunity is coordinated through a complex network of intercellular communication. Similar to the sculpting of tumor antigens during oncogenesis, a related hypothesis is that proteins secreted by malignant cells are shaped by somatic evolution. Malignant cells that then emerge secrete proteins that alter intercellular communication to promote tumor growth. As exosomes represent an emerging mode of cell-to-cell communication through the delivery of proteins and coding and non-coding RNAs, the objective of this study was to test this hypothesis by characterizing the impact of exosomes derived from three melanocyte cell lines on T cell function, with an emphasis on exosomal mRNA.

O 1: The Effects of Vitamin D Supplementation on the T cell Compartment in Multiple Sclerosis

Multiple sclerosis (MS) is a complex neurological disease and its prevalence is about 2 million in the world. Neuroinflammation plays a key role in MS. Vitamins are essential nutrients that have effective role on immune system including activation of lymphocyte and differentiation of T-helper cell. Vitamin D is a micronutrient that is effective on immune function. Deficiently of Vitamin D is a ...

Synthesis and Preclinical Characterization of [18F]FPBZA: A Novel PET Probe for Melanoma

INTRODUCTION Benzamide can specifically bind to melanoma cells. A 18F-labeled benzamide derivative, [18F]N-(2-diethylaminoethyl)-4-[2-(2-(2-fluoroethoxy) ethoxy)ethoxy]benzamide ([18F]FPBZA), was developed as a promising PET probe for primary and metastatic melanoma. METHODS [18F]FPBZA was synthesized via a one-step radiofluorination in this study. The specific uptake of [18F]FPBZA was studie...

Immunoaffinity-based isolation of melanoma cell-derived exosomes from plasma of patients with melanoma

Tumour-derived exosomes (TEX) are a subset of extracellular vesicles (EVs) present in body fluids of patients with cancer. The role of this exosome subset in melanoma progression has been of interest ever since ex vivo studies of exosomes produced by melanoma cell lines were shown to suppress anti-melanoma immune responses. To study the impact of melanoma-derived exosomes (MTEX) present in pati...

Exosomal cancer immunotherapy is independent of MHC molecules on exosomes

Peptide-loaded exosomes are promising cancer treatment vehicles; however, moderate T cell responses in human clinical trials indicate a need to further understand exosome-induced immunity. We previously demonstrated that antigen-loaded exosomes carry whole protein antigens and require B cells for inducing antigen-specific T cells. Therefore, we investigated the relative importance of exosomal m...

Cytotoxic T lymphocyte antigen-4 and immune checkpoint blockade.

The relationship between cancer and the immune system is complex and provides unique therapeutic opportunities. Cytotoxic T lymphocyte antigen-4 (CTLA-4) is a regulatory molecule that suppresses T cell effector function following initial activation by costimulatory signals. Fully human monoclonal antibodies targeting CTLA-4 have been shown to increase T cell function and antitumor responses in ...
